1177 Treatment of mice with mutant p53 peptide results in a cell-mediated immune response

Chronic, excessive exposure to solar ultraviolet radiation is the major causative agent for cutaneous squamous cell carcinomas. Mutations in p53 are essential these cancers develop. UV also immunosuppressive and an additional requirement UV-induced carcinomas The objective of this study determine whether a mediated immune response develops mutations p53. To accomplish our objective, we used immunization procedure using peptides that contain mutation commonly found keratinocytes following exposure. Cohorts C3H/HeN mice were injected subcutaneously with mutant peptide on their abdomen. Five days later, same was ear mice. No significant change thickness observed over 72 h period post injection. However, when anti-CD4 antibody (100ug/mouse) three prior first injection, there increase (p<0.05) cell-mediated Animals pretreated isotype control instead anti-CD4, did not have response. Immunization wildtype produce or without treatment anti-CD4+ antibodies. These findings indicate generated masked by regulatory CD4+T-cells.
